Simpsons, by a mile. Homer Simpson is the greatest cartoon character ever created IMHO.

Simpsons without a doubt, no matter how many times you've seen them they still make you laugh, and that's the mark of class.

It certainly true that the old ones can still make me laugh even after seeing them for the nth time, but with the new ones I struggle to laugh even on the first viewing. It's a sad state of affairs And if they had given the Simpsons time slot to either Futurama or Family Guy they both would have done much better (although supposedly Fox thought Family Guy was too edgy for them so they might have still cancelled it even if it was doing well).

Due to the joys of the massive American cable TV networks I watched a rerun of the Simpsons (a fairly recent episode), Family Guy and Futurama last night and I think head-to-head Family Guy was still the funniest.
