I don’t whether this is the same film but there was definitely a “sword and sandals” epic with John Wayne in it as a centurion (?).

At Christ’s crucifixion Wayne spoke “Truly, he is the son of God.” The director was not happy with it and said “Hey John, say it with more awe.”

Wayne obliged - “Awww! Truly, he is the son of God.”
