I rather suspect that many of the selections that folk make for a thread like this will be more lead by emotions than a feeling of what would constitute the best number one. I think in reality there cannot be a 'best' because the music covers so many genres. That said, I'm enjoying the selections, whatever the reasons for them.
That's why I could never answer the op's question. So many different songs have various emotional meanings to me across a lot of genres, it would be impossible to pick one. It would take a big effort to get it under a top 50.

Not THE greatest song in the world, technically, lyrically or musically, but my greatest song to raise my mood, invigorate me and get me off of my arse and pushing forward with what needs doing.

The vibe i get from let down is a loss of hope and people. It is certainly very powerful

Quite possibly this one. Structurally superb it has a unique intro, verse-pre-chorus-chorus sections, a bridge, and a long instrumental section. In particular the bridge breakdown is superb with the two part harmony. What also takes it to the next level is the extensive use of down picking by the iron wrist of James Hetfield. It just doesn't sound the same with alternate picking.

The version below is even more astonishing as it's played in a monsoon at Citeh's Etihad stadium and barely a bum note in sight.

Ooooh, this is a great thread. So much joy packed away in here.

Of those mentioned already, I love Echoes and Five Years. Just adore them.

But for me, and this is a purely personal thing, it has to be this. Been lucky enough to see it live three times, the last two with my daughter, who was only two years old when it was released and used to have to put up with me playing it far too loud in the car while I drove her round the streets of Delhi. Each time I’ve seen it, I’ve been on the spectrum between light weep and sobbing.

Already got my ticket to see it a fourth time at Sheffield at Tramlines this year.

Earmarked as my funeral song, which isn’t morbid at all. It’s perfectly thought out. It’s never played on the radio EVER. You have to seek it out, which means it will never sneak up on unsuspecting souls and upset them after I’m gone.

I have no idea how many times I’ve heard this song, must be somewhere in the thousands but it still sounds as fresh to me as it did when I first heard it. Most songs bore me to tears when I‘ve heard them repeatedly, not this one

This possibly my favourite version based on how many of my musical heroes are on it
